If Grow Tapped Card is on, the Freecell or Column you tap on the first tap of a move grows slightly, to indicate that it is selected.
If Tint Tapped Card is on, the Freecell or Column you tap on the first tap of a move is tinted slightly, to indicate that it is selected.
If Highlight Destinations is on, then after the first tap of a move, selecting a Freecell or Column you want to move from, all Foundations, Freecells, and Columns are dimmed, except those that you can legally move to — thus showing where you can perform the second tap.
(The meaning of “legally” depends on your Sequence Moves and Supermoves settings.)
In this layout, I’ve tapped on Column 1, and Sequence Moves is on, so we can move to Column 2. Highlight Destinations is on, so Column 2 is undimmed:
WARNING: If you turn off Grow Tapped Card and Tint Tapped Card and Highlight Destinations, there will be no indication of what was selected by the first tap of a move, or even that you have performed the first tap and that TapTapFreeCell is waiting for the second tap. That’s not a good interface! You can do that, but don’t.
